چکیده

Introduction: Cervical screening would benefit from a test based on a disease-specific biomarker that identifies high grade lesions, which could also indicate the presence of early precancerous lesions that have a high risk of progression to cancer. One such potential biomarker is p16INK4A. Objective of the study was to study the biomarker p16INK4A expression by immunostaining in cervical intraepithelial neoplasia. Material and method: Experimental study conducted from November 2009 to April 2011. 1500 women were screened for cancer cervix using conventional Pap test, VIA and VILI. Women having positive results underwent colposcopy and biopsy if required. P16INK4A expression in biopsy samples was studied using immunohistochemistry. Results: All test positive cases n= 235, underwent colposcopy. Colposcopic abnormalities were detected in n=83 and biopsy proven cervical intraepithelial neoplasia(CIN) in n=15. P16INK4A expression was seen in eight of 15 CIN cases. The strength of positivity of P16INK4A was higher in CIN 2 and CIN 3 cases as compared to CIN 1. Also, the pattern of staining differed among various grades of CIN. Conclusion: P16INK4A expression was seen in majority of CIN 2 and CIN 3 lesions suggesting a higher grade lesion as compared to CIN 1. The discrimination between the lesions can be made without having to depend on traditional histopathology which gives a static picture of pre-invasive lesions of the cervix. This can help in differentiating high grade lesions from the lower grades, thereby helping in appropriate management

Biomarker expression in cervical intraepithelial neoplasia: potential progression predictive factors for low-grade lesions.

The aim of this study was to reveal whether 3 biomarkers (p16INK4a, ProEx C, and human papilloma virus DNA) are useful in the diagnosis of cervical intraepithelial neoplasia and whether they could predict disease progression of cervical intraepithelial neoplasia-1. We analyzed 252 cervical specimens: nondysplastic mucosa (n = 9), cervical intraepithelial neoplasia (n = 229), and squamous cell c...

Evaluation of Ki67, p16 and CK17 Markers in Differentiating Cervical Intraepithelial Neoplasia and Benign Lesions

Background: Cervical intraepithelial neoplasia (CIN) is a premalignant lesion capable of progressing to cervical cancer. Despite the existing well-defined criteria, the histomorphologic diagnosis is subject to high rates of discordance among pathologists. The aim of this study was to evaluate Ki-67 (MIB-1), CK17 and p16 INK4a (p16) markers by immunohistochemical methods in differentiating CIN f...

Liquid-based cytology--new possibilities in the diagnosis of cervical lesions.

Liquid-based cytology (LBC) enables the use of supplementary methods in the diagnosis and prognosis of cervical lesions. The aim of this study was to analyze the correlation between p16-INK4a immunoexpression in ThinPrep cervical cytologic samples and human papillomavirus (HPV) detection by polymerase chain reaction (PCR) from the same sample. LBC-ThinPrep (Cytyc, USA) cervical cytology samples...

p16ink4a is a surrogate marker for high-risk and malignant cervical lesions in the presence of human papillomavirus.

OBJECTIVE To strengthen the role of high-risk human papillomaviruses (HPVs) in the development of cervical cancer, and the association between HPV and the cell cycle inhibitor gene p16ink4a in cervical carcinogenesis. METHODS In this study, the association between p16ink4a and the presence of HPV DNA in cervical lesions was investigated in a total of 177 cervical biopsies classified as benign...
